Preparation

  • Shake the container of Demi Fraser Broth Base well.
  • Suspend 57.4 g in 1 L of distilled water.
  • Autoclave at 121°C for 15 minutes.
  • Cool to 50°C
  • Aseptically add the contents of proper amount of Demi Fraser Selective Supplement (10 mg Nalidixic Acid and 12.5 mg Acriflavine Hydrochloride) and Ferric ammonium citrate (0.5 gr).

STORAGE

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Dehydrated Appearance: Free flowing, homogeneous, tan.
  • Prepared Appearance: Medium amber, clear to slightly opalescent with a fine precipitate.
  • Inoculate and incubate at 35 ± 2°C for 24 – 48 hours.
