SIMMONS Citrate Agar

Application: For the differentiation of Gram-negative bacteria on the basis of citrate utilization. Bacteria that can utilize citrate as sole carbon source turn the medium blue.

Preparation

  • Shake the container well to homogenize the powder. Suspend 22.3 g of the powder in 1 L of purified water. Mix thoroughly. Before using distilled water, check the pH level of the water to be between 6.5-7. If this point is not considered, the original color of the medium may be different.
  • Mix thoroughly. Heat with frequent agitation and boil for 1 minute to completely dissolve the powder.
  • Autoclave at 121°C for 15 minutes.
  • Allow to cool in a slanted position for use as slants. The agar also may be used as a plating medium.

STORAGE

Store dehydrated medium below 30 °C and prepared medium at 2-8 °C. The best time to use is before the expiration date printed on the product label.

Quality Control

  • Dehydrated Appearance: Light orange free-flowing, homogeneous may contain many dark and gray flecks.
  • Prepared Appearance: Medium to dark, green, clear to slightly hazy, with a small amount of precipitate and as many as a large amount of insolubles.
  • Cultural Response after inoculation with fresh cultures and incubate aerobically at 35 ± 2°C for 24-48 hours.
