Mitis Salivarius Agar

Application: For the isolation of Streptococcus mitis, Streptococcus salivarius, and Enterococci, particularly from heavily contaminated samples, use a 1% potassium tellurite (iS47083) solution.

Preparation

  • Shake the container well to homogenize the powder. Suspend 90 g of the powder in 1 L of purified water.
  • Autoclave at 121°C for 15 minutes.
  • Let the medium cool to a temperature of 55-50 degrees Celsius, then add 1 milliliter of 1% potassium tellurite solution (iS47083).
  • After adding potassium tellurite solution, do not heat the medium under any circumstances.

 

STORAGE

Store dehydrated medium below 30 °C and prepared medium at 2-8 °C. The best time to use is before the expiration date printed on the product label.

 

 

Test

Test

Immediately after reaching the laboratory, inoculate the sample on the medium.
Incubate the plates at 35 ± 2 °C for 24-48 hours and 5% carbon dioxide.
Also culture a non-selective plate (such as blood agar) from the same sample. This increases the chance of recovery of microorganisms that are in small numbers in the sample. It is also an indicator for comparison with Mitis salivarius and the presence of other microorganisms in the sample.

  • Prepared Appearance: Deep royal blue, slightly opalescent
  • Cultural response was observed under 5-10% CO2 at 35 ± 2°C for 18-48 hours.
