Preparation

  • Shake the container well to homogenize the powder. Suspend 20.38 g of the powder in 1 liter of distilled or deionized water. Mix well.
  • Add enough amount to the test tube.
  • Autoclave at 121°C for 15 minutes.

 

STORAGE:

Store dehydrated medium at 10-30 °C and prepared medium at 10-25 °C. The best time to use is before the expiration date printed on the product label.

Glucose is the most important carbohydrate used in OF Basal culture medium. Glucose OF culture medium is also recommended by the ISO committee.

Carbohydrate to acid fermentation is indicated by the bromothymol blue pH indicator with a color change from blue to yellow.

If the environment is exposed to air oxygen, oxidative utilization occurs in the culture medium and results in a change in the color of it.

It is necessary to cover the surface of the culture medium with mineral oil.

The low amount of agar in the culture medium helps to detect motility.

It is necessary to do each inoculation in pairs. Cover the culture medium in one tube with mineral oil and leave the other uncovered. Microorganisms perform fermentation, showing acid production in both tubes. Oxidative microorganisms show an acid production reaction in the uncoated medium and also a slight growth without causing any change in the coated medium.

Microorganisms that are not classified in any of the oxidative or fermentative categories do not show any change in the coated medium. But in the uncovered medium, they are characterized by an alkaline reaction. The acidic reaction of oxidative organisms appears on the surface of the culture medium and gradually spreads. If the oxidative reaction is slow, an initial reaction on the surface of the uncoated tube may persist for several days and eventually change to an acidic reaction.
