Application: Thermoacidurans agar culture medium is used to culture and isolate the bacterium Bacillus thermoacidurans (Bacillus coagulans) from food materials.

Preparation

  • Shake the container containing the Themoacidurans Agar culture medium powder well. Dissolve 39.2 grams of the medium in 1 liter of distilled water.
  • Boil until it is completely dissolved.
  • Autoclave at 121 degrees Celsius for 15 minutes.

STORAGE

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

Test

  • Heat-shock the desired sample and add 1 milliliter of it to the plate.
  • Add 15-20 milliliters of cooled culture medium (45-50 degrees Celsius) to the plate.
  • Gently rotate the plate to mix the sample and the medium well.
  • Incubate the plates at 55±1°C for 3-5 days.

Results and Interpretation

After incubation, examine the plates for colony morphology and number.

Note

  • Other bacteria may also grow on this medium besides Bacillus themoacidurans. To ensure the type of bacteria, perform microscopic and biochemical tests.
