Orange Serum Agar OSA

Application: This medium is used for identification, cultivation, and enumeration of acid-resistant and food spoilage microorganisms, especially in fruits.

Preparation

  • Shake the container well. Dissolve 42 grams of the medium in 1 liter of distilled water.
  • Heat, if necessary, to facilitate dissolution.
  • Autoclave at 115°C for 15 minutes. Avoid overheating the medium.

STORAGE

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • For the total count of acid-resistant microorganisms by the pour-plate method, dilute the sample 10-fold and add 1 ml to the plate.
  • Add 15-20 ml of cooled (45-50°C) culture medium to the plate.
  • Gently rotate the plate to mix the sample and the medium well.
  • Incubate the plates at 30±2°C for 3-5 days.

Results and Interpretation

After incubation, examine the plates for colony morphology and number. Since Orange Serum Agar is not a selective medium, microscopic examination and biochemical tests are required for precise identification of bacterial species and genus.

Note

  • Avoid overheating this medium as it may cause darkening of the color and loss of its solidification properties.

  • Dehydrated Appearance: Fine, homogeneous, free of extraneous material, may contain dark tan particles.
  • Prepared Appearance: Light to medium, yellow to tan; clear to slightly hazy
  • Cultural response after incubation the inoculated culture medium for up to 4 days at 28 °C aerobically. if fungi are suspected to be present, incubate for up to 5 days. Further tests can be performed to differentiate and identify the colonies.
