Preparation
- Thoroughly shake the container. Dissolve 20 grams of the medium powder in one liter of distilled water.
- Heat the solution gently and boil for a few seconds until the medium is completely dissolved.
- Autoclave the culture medium at 121 degrees Celsius for 15 minutes.
STORAGE
The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

- Before inoculating, plates should be placed in the laboratory temperature to equilibrate with the room temperature.
- If the sample is transferred to the culture medium using a swab, the swab should be streaked on a corner of the plate. Then, this area is streaked several times to obtain single colonies.
- In addition to this culture medium, selective culture media can also be used.
- Incubate the plates at 35-37°C for 18-48 hours.
Results and Interpretation
After incubation, check the plates for colony growth.
Note
- Nutrient Agar culture medium contains 1.5% sodium chloride (NaCl) and is more alkaline than this medium. Adding 0.8% NaCl to the medium helps maintain osmotic balance and prevents cell lysis.
- The Nutrient Agar culture medium prepared in the tube is used for the cultivation and maintenance of pure cultures. The bacteria cultured in the tube can be used for serological and biochemical tests.
