Preparation

  • Shake the container well. Dissolve 21.5 grams of the powder medium in 450 milliliters of distilled water. Heat gently until the medium is completely dissolved.
  • Autoclave at 121 degrees Celsius for 15 minutes.
  • After the medium cools and its temperature reaches 45-50 degrees Celsius, dissolve the content of 1 vial of Bacillus cereus supplement (iS47004) in 5 milliliters of distilled water and add it to the medium after sterilization by 0.22-micrometer filter.
  • Add 50 milliliters of egg yolk emulsion (iS47020) to the medium in a sterile manner.
  • Mix the medium well and pour it into the plate.

STORAGE

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Before inoculation, dry the surface of the culture plate.
  • Dilute the food sample in a suitable buffer or peptone water medium (i23137).
  • Spread approximately 0.1 milliliters of the diluted sample with a sterile spreader on the surface of the plate.
  • Incubate the plates at 30±2 degrees Celsius for 18-40 hours.

Results and Interpretation

Bacillus cereus colonies on MYP Agar appear pink/purple, rough and dry with a halo of dense precipitate (caused by the bacterial lecithinase enzyme on egg yolk lecithin) surrounding them. For counting bacterial colonies, colonies with these characteristics are used. Colonies with no halo or those surrounded by a colorless or yellow halo are not Bacillus cereus.
