Fluid Thioglycollate Medium

Application: This medium is suitable for culturing obligate anaerobic bacteria, facultative anaerobic bacteria, oxygen-tolerant bacteria, as well as performing sterility tests.

  • Shake the container well. Dissolve 29.8 grams of the medium powder in 1 liter of distilled water.
  • Heat and stir gently until the powder is completely dissolved.
  • Pour the medium into test tubes in volumes of 10 milliliters.
  • Autoclave at 121 degrees Celsius for 15 minutes.

STORAGE:

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Insert the specimen to be examined into the medium. Try to inoculate the specimen at the bottom of the tube.
  • To ensure anaerobic conditions for culturing, 1 milliliter of paraffin or sterile agar solution can be added to the surface of the medium in the tube.
  • Incubate the cultured specimen for up to 5 days at a temperature of 35-37 degrees Celsius.

Results and Interpretation

Turbidity in the lower part of the tube indicates the growth of anaerobic bacteria. Bacteria that are obligate aerobes grow in the oxidized portion of the tube at the top. Obligate anaerobes only grow in the bottom part of the tube, which is located under the oxidized part.

Note

  • Always use fresh Fluid Thioglycollate culture medium.
  • Do not use the medium if more than one-third of the upper part of the tube is pink. In this case, heat the medium to remove the oxygen inside. If after reheating, a large portion of the medium remains pink, do not use it.
  • Do not heat the medium more than once as it can become toxic.
  • It is possible that the growth of obligate anaerobic bacteria is limited due to the rapid growth of facultative anaerobic bacteria. Therefore, if no growth is observed, further testing and Gram staining should be performed.
