Corn Meal Agar

Application: This is a multi-purpose medium for cultivating fungi. Poly-sorbate 80 is also used to test the ability of Candida strains to produce chlamydospores.

Preparation

  • Shake the container containing the cornmeal agar culture medium powder well. Dissolve 17 grams of the culture medium powder in 1 liter of distilled water.
  • If necessary, add 1% poly-sorbate 80 (i880218) or 1% dextrose (i880081) and mix well.
  • Heat for 1 minute until the powder is completely dissolved in water.
  • Autoclave at 121°C for 15 minutes.

STORAGE:

Store dehydrated medium below 30 °C and prepared medium at 2-8 °C. The best time to use is before the expiration date printed on the product label.

  • Take a colony from the surface of the SDA medium and create a deep groove in the cornmeal agar medium, like a horizontal slot.
  • Repeat this process for each colony.

Results and Interpretation

  • After 24-48 hours of incubation at a temperature of 25 ± 30 degrees Celsius, growth should be examined using a microscope.
  • Candida albicans produces clusters of round balls with mycelium of germinating cells and thick-walled chlamydospores.

Note

  • Organisms must be isolated in a suitable culture medium before cultivation for identification.
  • A cornmeal agar plate can be used to identify four or five different colonies of Candida that have grown on SDA.
  • Adding 0.001% (w/v) trypan blue to the culture medium provides a suitable background for better observation of morphological features.
