Dichloran Rose Bengal Chloramphenicol (DRBC) Agar

Application: This is a selective medium for enumeration of food spoilage fungi and yeast.

  • Thoroughly shake the DRBC Agar culture medium container. Dissolve 31.6 grams of the medium in 1 liter of distilled water.
  • Boil the solution until the medium powder is completely dissolved in water.
  • Autoclave the medium at 121 degrees Celsius for 15 minutes.
  • After autoclaving, cool the medium to 50 degrees Celsius and then pour it into the culture plate.

STORAGE:

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Dilute the desired food sample according to the relevant protocol and prepare a serial dilution from it.
  • Place 0.1 mL of the sample on the agar plate and spread it completely over the surface.
  • Incubate inoculated plates in 25 degrees Celesius up to 5 days.

Results and Interpretation

  • Check the growth on the plate after 3, 4, and 4 days of incubation at 25 degrees Celsius.
  • Yeasts colonies on this culture medium appear pink due to the absorption of rose bengal.
  • Calculate the number of colonies per 1 gram of food.

 

Note

  • The DRBC culture medium has a low pH which can increase the likelihood of yeast growth inhibition in the presence of rose bengal. For this reason, if it is necessary to count yeast in addition to mold, it is better to use both Rose Bengal Chloramphenicol Agar (Catalog number i23159) and DRBC culture medium for culturing the sample simultaneously.
  • Due to the unique properties of DRBC Agar, it is possible that some fungal or yeast strains may not grow or show poor growth on this culture medium. Also, it is possible that some bacteria may not be completely or relatively inhibited from growing on this medium.
  • Keep in mind that DRBC Agar should not be exposed to light, as the rose bengal present in this medium is oxidized in the presence of light and produces toxic substances that are harmful to mold and yeast growth.
