Application: for identification of microbial DNase enzyme especially in Staphylococcus.

Preparation

  • Shake the container well to homogenize the powder. Weigh 42 grams of the powder and dissolve it in 1 liter of distilled water.
  • Heat it gently until the medium is completely dissolved.
  • Autoclave in 121 degree Celsius for 15 minutes.
  • Allow it to cool down to a temperature of 50 degrees Celsius and then pour in strile petri disehs.
  • After culturing and growing bacteria, to determine the result of the DNase test and halo formation, use Hydrochloric acid (i880090) with concentration of 1 N to cover the surface of the plate.

 

STORAGE:

Store dehydrated medium below 30 °C and prepared medium at 2-8 °C. The best time to use is before the expiration date printed on the product label.

  • Dehydrated Appearance: Light-beige, free-flowing, homogeneous
  • Prepared Appearance: Light to medium amber, slightly opalescent, may have a slight precipitate.
  • Cultural response was observed after up to 48 hours of incubation at 35 ± 2°C. After incubation carefully flood the surface of the plates with 1 N hydrochloric acid and check the zone of colonies.
