EC broth (medium)

Application: Used for the specific identification of Coliforms at 37°C and E. coli at 44.5°C in water, food, and other substances.

Preparation

Shake the container well. Dissolve 37 grams of the medium powder in 1 liter of distilled water.
Heat gently to dissolve completely.
Pour into test tubes containing Durham tubes.
Autoclave at 121 degrees Celsius for 15 minutes.

STORAGE:

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

Test

Inoculate the desired sample into a test tube containing the culture medium.
Incubate at 37°C for 48-24 hours to examine Coliforms and at 44.5°C for 24-48 hours to examine E. coli.

Results and Interpretation

Bacterial growth (indicated by turbidity in the medium) along with gas production is a positive indication of the presence of enteric bacteria in the sample.

Note

If using water samples that are acidic, contain antimicrobial substances, or have been stored in the refrigerator, false-negative results for the growth of Coliforms may occur.
