Giolitti-Cantoni Broth Base

Application: Giolitti-Cantoni Broth Base is used for enriching Staphylococcus aureus from foods during isolation procedures.

Preparation

  • Shake the container well to homogenize the powder. Dissolve 55.2 g of the powder in 1 L of purified water.
  • Dispense 19 mL amounts into 20 × 200 mm tubes.
  • Autoclave at 121°C for 15 minutes.
  • Cool to 15-30°C.
  • Add 1.05 ml of 1% potassium tellurite solution to each test tube under sterile conditions. If the sample under test is meat (or quality control microorganisms), add 0.105 mL of 1% potassium tellurite solution (iS47083) to each test tube.
  • Mix well. Inoculate test sample to the test tubes.

 

STORAGE:

Store dehydrated medium below 30 °C and prepared medium at 2-8 °C. The best time to use is before the expiration date printed on the product label.

Testing process

Giolitti-Canton medium is used to identify Staphylococcus aureus in Skim Milk or other baby food, which should not contain the amount of microorganism in 1 gram. This Culture medium is also recommended by the ISO committee for the examination of meat and meat products.
The culture medium should be inoculated as soon as possible after sterilization. Otherwise, it is necessary to remove the oxygen absorbed by the environment by placing the test tubes in flowing steam for 15-20 minutes.
Inoculate 1 g of sample or 1 ml of diluted sample in 19 ml of medium. Do 2 replicates for each sample.
Cover the medium with 5 ml of sterile melted paraffin and incubate at 37°C for 24-48 hours and check daily.
Blackening of the medium that normally occurs at the bottom of the tube within 48 hours indicates the presence of Staphylococcus aureus.
Inoculation of Baird-Parker agar medium using blackened Giolitti-Cantoni medium shows black colonies surrounded by a light halo.

Notes

Lithium chloride in the culture medium powder is extremely harmful. Avoid contact with the skin or inhalation of the powder. In case of skin contact, wash immediately with plenty of water.
