Preparation

Thoroughly shake the glucose agar container. Dissolve 41.52 grams of the medium in 1 liter of distilled water.
Heat until boiling and completely dissolve the medium.
Autoclave at 121 degrees Celsius for 15 minutes.
Pour the medium into screw-capped tubes.

STORAGE:

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

Test

Inoculate a suspected colony of Enterobacteriaceae, taken from a selective medium and purified in Nutrient Agar medium, into a deep layer of glucose agar using a culture needle.
Incubate at 35±2°C for 18-24 hours.

If Enterobacteriaceae is present in the medium and glucose fermentation occurs, acid is produced and the color of the medium at the end of the tube changes from purple to yellow.
