Application: This is a selective medium for the isolation and enumeration of pathogenic staphylococci from food samples, clinical specimens, and other materials.

Preparation

  • Shake the container well. Dissolve 108 grams of the medium powder in 1 liter of distilled water.
  • Heat the solution gently and boil for a few seconds until the medium is completely dissolved.
  • Autoclave the culture medium for 15 minutes at 121 degrees Celsius.

STORAGE

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Inoculate the sample onto the medium. Due to the high inhibitory properties of this culture medium, the sample should be inoculated in a large volume.
  • Incubate the plates at 35 degrees Celsius for 3 days.

Results and Interpretation

Bacteria that can grow in the presence of high levels of salt and ferment manitol in this medium produce acid, which causes the pH indicator, phenol red, to turn from red to yellow. Therefore, coagulase-positive staphylococci produce yellow colonies with a yellow halo, while coagulase-negative staphylococci produce small, red colonies without any color change in the culture medium.

Micrococcus will be observed as large colonies, white to orange in color, without any color change in the culture medium. Most other bacteria will be inhibited in this culture medium. The growth of most other bacteria will be inhibited in this culture medium.

  • Cultural Response after inoculation and incubate at 35 ± 2°C for 42-48 hours. Incubate plates with Staphylococcus aureus ATCC 6538 and E. coli ATCC 8739 at 30-35°C for 18-72 hours.
