Mueller Hinton Agar

Application: Mueller Hinton Agar is recommended for antimicrobial disc diffusion susceptibility testing of common, rapidly growing bacteria by the Bauer-Kirby method, as standardized by the CLSI.

Preparation

  • Shake the container well to homogenize the powder. Suspend 34 g of the powder in 1 L of purified water.
  • Autoclave at 121°C for 15 minutes.
  • Avoid overheating.
  • OPTIONAL: Cool medium to 45-50°C and aseptically add 5% sterile defibrinated sheep blood.
  • Pour cooled Mueller Hinton agar into sterile Petri dishes on a level, horizontal surface to give a uniform depth of about 4 mm and cool to room temperature.

STORAGE

Store dehydrated medium below 30 °C and prepared medium at 2-8 °C. The best time to use is before the expiration date printed on the product label.

Use this medium only for pure cultures.
The amount of cultured bacteria on the culture medium affects the size of the surrounding halo. If a high amount of bacteria is cultured on the medium, smaller halos are formed, and if the amount of inoculated bacteria is very low, the created halo will be larger.
Fastidious organisms will not grow on this medium and need to be supplemented with blood.
Fastidious anaerobes will not grow on this medium.
Antimicrobial sensitivity is done with antibiotic discs. The storage conditions of the disks must be suitable. Improper storage conditions of discs can affect their antibiotic power.
In certain circumstances, antibiotic susceptibility results in vitro will not be the same as those seen in vivo.

  • Prepared Appearance: Light to medium amber, slightly opalescent.
  • Prepare the medium per label directions. Using the organisms listed below, inoculate plates, add antibiotic disks and incubate as recommended by CLSI. Measure zone diameters and compare to the CLSI recommended zone ranges.
