PA Broth

Application: Used for detecting coliforms in treated water.

Preparation

Shake the container well to homogenize the powder. Suspend 91.5 g of the powder in 1 litre of purified water for the preparation of the triple strength concentrated broth.
Mix thoroughly. Warm gently to completely dissolve the powder.
Dispense 50 mL amounts into screw cap 250 mL milk dilution bottles.
Autoclave at 121°C for 12 minutes.

STORAGE

Store dehydrated medium below 30 °C and prepared medium at 2-8 °C. The best time to use is before the expiration date printed on the product label.

Quality Control

Dehydrated Appearance: Beige, free-flowing, homogeneous.
Prepared Appearance: Purple, clear to very slightly opalescent, without significant precipitate.
Prepare the PA Broth medium in triple strength. Sterilize 50 mL of the medium in 250 mL sterile flasks. Add 100 mL of drinking water after medium is sterilized and cooled to room temperature. Inoculate bottles with the test organisms. Cultural Responce after incubation bottles at 35 ± 0.5°C for 18-48 hours.
