Preparation

  • Shake the container of Rappaport Vassiliadis Broth well.
  • Suspend 26.8 g of the medium in 1 L of purified water.
  • Dispense 10 mL into glass tubes.
  • Dispense 10 mL into glass tubes, cap and autoclave at 115°C for 15 minutes.

STORAGE

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Dehydrated Appearance: Pale green to green, free-flowing, homogeneous.
  • Prepared Appearance: Dark blue, clear.
  • Inoculate the culture medium with the sample or material from a pre-enriched culture (e.g., Buffered Peptone Water) and incubate for 24 hours at 41.5 °C. Streak material from the resulting cultures onto selective culture media.
