Violet Red Bile Glucose Agar (VRBGA)

Application: Isolation and enumeration of enterobacteria in food samples.

Preparation

  • Shake the container of the VRBGA medium well. Dissolve 39.5 grams of the medium in 1 liter of distilled water.
  • Heat it until boiling and completely dissolved. Do not heat it for more than 2 minutes after that.
  • Do not autoclave the medium. Avoid overheating the medium.
  • After the medium has cooled, mix it well and pour it into the petri dishes.
  • Prepared plates can be stored for up to 2 weeks at 4°C, away from light.

STORAGE

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • This medium is used in both the pour plate (with or without overlay) and spread methods.
  • Inoculate the sample into the culture medium according to your desired standard method.
  • Incubate at 35±2°C for 18-24 hours.

Results and Interpretation

Red colonies that have a dark halo (due to the precipitation of bile salts) around them are likely to be enterobacteria. For confirmation, select suspicious colonies for fermentation and oxidase tests.
